ABSTRACT

Objective:To explore the association of cardiometabolic index(CMI) and other body fat evaluation indicators [body mass index(BMI), waist circumference(WC), waist to height ratio(WHtR), lipid accumulation index(LAP)] with the prevalence of metabolic syndrome(MS) as well as the predictive value of the above indicators for MS.Methods:A total of 10 140 residents over 40 years old in Guiyang city who participated in the " Epidemiological study on tumor risk of type 2 diabetes patients in China" in 2011 were recruited. The 2005 International Diabetes Federation diagnostic criteria were used to identify MS. Logistic regression was used to analyze the association of CMI and other body fat evaluation indicators with MS. Receiver operating characteristic(ROC) curve was used to evaluate the predictive value and the optimal cut-off point of different indicators. Taking the best cut-off point value of each index as the boundary, the prevalence of MS was evaluated again by Chi square test.Results:The prevalence of MS in the study population was 39.81%(27.23% for men and 44.39% for women). Logistic regression analysis showed that the risk of MS increased with increasing CMI and quartile level of other body fat evaluation indicators in both men and women( P<0.05). The risks of MS in CMI Q4 group were 17.15(95% CI 11.64-25.27) for male and 45.14(95% CI 37.07-54.96) for female compared with Q1 group. In male, the area under curve(AUC) of MS by predicted CMI was 0.761(sensitivity 79.8%, specificity 63.2%, optimal cut-off point 0.71). WC displayed the highest value of AUC among the body fat evaluation indicators. In women, the AUC value of MS predicted by CMI was 0.831(sensitivity 76.8%, specificity 75.7%, optimal cut-off point 0.65), higher than those of BMI and WHtR while lower than those of WC and LAP. Further calculating the prevalence of MS with the best cut-off point value of each index as the boundary, WC was still the best predictor for male, while CMI was only secondary to LAP for women. Conclusion:CMI and other body fat evaluation indicators are significantly associated with MS. CMI could be used to predict MS.

ABSTRACT

Objective:To investigate the relationship between cardiometabolic index and metabolic syndrome in people aged 40 and beyond in Guiyang city.Methods:A total of 4 506 residents over 40 years(including 3 067 females and 1 439 males) were enrolled in the analysis from those who participated in the epidemiological study of cancer risk in Chinese patients with type 2 diabetes in 2011 in Guiyang City. The cardiometabolic index (CMI) is calculated by triglycerides(TG)/high-density lipoprotein-cholesterol(HDL-C)×waist-to-height ratio (WHtR). Multivariable logistic regression was used to analyze the correlation between cardiometabolic index and metabolic syndrome, and ROC was used to analyze the predictive ability of CMI on the incidence of metabolic syndrome. Results:The average follow-up period was 3 years. According to the International Diabetes Federation (IDF) diagnostic criteria for metabolic syndrome in 2005, 985 patients (774 women and 211 males) had metabolic syndrome. The incidence rate of metabolic syndrome in the general population was 21.86%, the incidence rate of male metabolic syndrome was 14.66%, and that of women was 25.24%, and the incidence of CMI increased with the increase of the number of women. After multivariable logistic regression analysis, the odds ratio of CMI for metabolic syndrome in women is 1.303(95% CI 1.263-1.344) and 1.724(95% CI 1.162-2.558) in men, respectively. ROC results showed that CMI had a good ability to predict the incidence (AUC: 0.759 for men and 0.852 for women). Conclusion:CMI is positively associated with the incidence of metabolic syndrome. It supports CMI as a useful method to screen metabolic syndrome in China′s general population.

ABSTRACT

Objective@#To explore the influence of metabolic syndrome on the risks of cardiovascular disease (CVD) events and all-cause mortality.@*Methods@#In this prospective cohort study, urban residents aged 40-79 years in Guiyang were followed-up for three years. The end-points were CVD events and all-cause mortality. COX proportional hazards model were used for the corresponding hazard ratios (HRs) of CVD events and all-cause mortality. Metabolic syndrome was defined according to the 2005 International Diabetes Federation (IDF) criteria.@*Results@#A total of 7 313 subjects were included. 146 cases of CVD events and 80 cases of all-cause mortality were recorded. After adjusting for gender, age, smoking, alcohol consumption, and blood lipid levels, the metabolic syndrome increased the risks of CVD events and all-cause mortality. The HRs were 1.43 (P=0.037) and 1.25 (P=0.418), respectively. As compared metabolic syndrome with non-metabolic syndrome, an increased risk of CVD events was found. The HR was 1.43 (P=0.013). No significantly increased risk of all-cause mortality was found in subjects with metabolic syndrome.@*Conclusion@#The metabolic syndrome was associated with increased risk of CVD events and there was no significant increase in all-cause mortality. Metabolic syndrome is an important risk factor for CVD events.

Objective To investigate the effect ofpcDNA3.1 (+)/VEGF165 plasmid transfection on endothelial progenitor cells (EPCs)derived from murine bone marrow mediated by liposome.Methods EPCs were isolated from murine bone marrow cuhured in vitro,identified by immunocytochemistry and fluorescence-activated cell sorting.After transfection,VEGF165 mRNA in EPCs was detected by reverse transcription polymerase chain reaction (RT-PCR),and VEGF protein in the culture supernatant of EPCs was detected by enzyme-linked immunosorbent assay (ELISA).MTT method was used to study the effect of transfection by pcDNA3.1 (+)/VEGF165 plasmid on EPCs.Results EPCs expressed cell markers CD34,CD133,KDR.Gel electrophoresis showed that a 576bp product was amplified by RT-PCR in VEGF165-transfected cells,while a weak expression of VEGF165 was observed in mock-transfected and non-transfected cells.The protein level of VEGF165 in the supernatant of VEGF165-transfected was higher than that in the other groups.VEGF165 plasmid transfer could stimulate EPCs proliferation.Conclusion EPCs can successfully transfect VEGF165 gene and promote the proliferation of EPCs,providing experimental basis for treatment of erectile dysfunction.

ABSTRACT

This paper presents a new pulse oximeter used to low perfusion at 0.125% and wide oxygen saturation range from 35% to 100%. In order to acquire the best PPG signals, the variable gain amplifier(VGA) is adopted in hardware. The self-developed auto-correlation modeling method is adopted in software and it can extract pulse wave from low perfusion signals and remove motion artifacts partly.

ABSTRACT

Objective To study the expression of pl6, FHIT genes in cervical carcinoma and its clinical significance. Methods By immunohistochemistry SP method, the expression of pl6, FHIT in different 118 cases of cervical lesions were detected and the results were analyzed in combination with clinical pathological features. Results Of 118 patients, 15 cases suffered cervicitis;38 cases took place cervical tumor-like changes;65 cases caught cervical cancer. p16 expression rates were 0, 33.3 %, 70.0 %, 87.5 %,and 92.3 % respectively;while FHIT expression rates were 73.3 %, 75.5 %, 60.0 %, 37.5%, and 30.8 % respectively. Compared with cervicitis, pl6 and FHIT expression rates in the cervix tumor-like changes,cervical carcinoma had significant difference (P <0.05). There was positive correlation in protein expression between p16 and FHIT (x2 =33.33, P <0.001). Conclusion Combination of p16, FHIT detection can be used as early diagnostic tool of cervical lesions and cervical cancer markers;meanwhile, the method can serve as a clinical evaluation of tumor biological behavior and prognosis of auxiliary indexes.

ABSTRACT

Objective To investigate the expression fragile histidinetriad (FHIT) protein in cervical carcinoma and its relationship with clinicopathological feature of the disease. Methods Immunohistochemistry SP was used to detect the expression of FHIT protein in 20 cases with chronic cervsis and 95 cases with Ⅰ aⅢ b stage cervical carcinoma before and after treatment. The association of the expression of FHIT with clinicopathological feature was analyzed by the statistical method. Results There were significant differences between FHIT expression and histological grades and types of tissue, lymph node metastasis and invade depth (P <0.05). FHIT expression was not correlated with age and clinical stage (P >0.05). There were significant differences in FHIT protein expression levels in the patients with cervial cancer between before-after radiotherapy and the levels before radiotherapy was lower then those after radiotherapy (P <0.05). There was positively correlated in FHIT protein expression rates before and after radiotherapy (P <0.05). There were significant correlation between FHIT expression and 3-year survival rate, the positive rates of the expression FHIT protein higher then negative ones (P <0.05). Conclusion FHIT protein has great reference value that could be as a parameter for evaluating biological action and predicting the prognosis of cervical cancer.

ABSTRACT

Objective:To explore the effect of radiotherapy on the FHIT protein expression and cell apoptosis of cervical squamous carcinoma and discuss the relationship between FHIT protein expression and cell apoptosis. Methods:Expression of FHIT protein was measured by immunohistochemical method and cell apoptosis was detected by TdT-mediated dUTP terminal nick end labeling (TUNEL) staining in 50 cases of squamous cell cervical carcinoma at ⅡB-ⅢB stages before, during (Dt 10 Gy and Dt 30 Gy), and after radiotherapy. Results:Of the 50 patients, the positive rates of the expression of FHIT protein was 56% at Dt 10 Gy, 68% at Dt 30 Gy, and 84% after radiotherapy, which were significantly increased compared with that before radiotherapy (36%, P<0.05). The positive rates of cell apoptosis was 52% at Dt 10 Gy, 64% at Dt 30 Gy and 78% after radiotherapy, which were significantly elevated compared with that before radiotherapy (28%, P<0.05). In the process of radiotherapy, cell apoptosis was positively related to the expression of FHIT protein (P<0.05). Conclusion:Radiotherapy reinforces the expression of FHIT protein and induces apoptosis cocurrently. FHIT protein has regulatory effects in cell apoptosis induced by radiotherapy.

BACKGROUND:A steady method that can isolate endothelial progenitor cells(EPCs) need to be explored.Studies show that the EPCs riches in bone marrow with strong reproductive activity,which can be easily obtained.OBJECTIVE:To evaluate the isolating culture and biological characterization of EPCs derived from rat marrow.DESIGN,TIME AND SETTING:The in vitro cytology experiment was performed at the central laboratory of the First Affiliated Hospital of Chongqing Medical University from August to December 2008.MATERIALS:Twenty SD rats,with two-week-old,were obtained from Experimental Animal Center of Chongqing Medical University.METHODS:femurs and tibias of rats were isolated under sterile conditions,and then the bone marrow cavity was washed by 4 ℃ 0.01 mol/L phosphate buffer(PBS).Mononuclear cells were collected from marrow suspension by density gradient centrifugation,followed by cultured with M199 culture medium containing 20% fetal bovine serum to obtained cell suspension.The cells were vaccinated into culture flask(coated with fibronectin) with the density of 1?109/L,and incubated at 37 ℃ in 5% CO2 atmosphere.MAIN OUTCOME MEASURES:Morphologic change of cells was observed by an invert microscope,the expression of CD133,CD34 as well as KDR was detected by immunocytochemistry,and the flow cytometry was used to analyze the growth curve.RESULTS:The adherent cells formed clusters that were found to be characterized by density packed round cells surrounded by spindle-likes cells.Adherent cells were identified positive for CD133,CD34 and KDR by immunocytochemistry on days 4,7,10 and 14.CD133-positive cells reduced after 10 days.Flow cytometry analysis indicated that the number of CD133/KDR double positive cells gradually increased to the maximum at day 10 and then declined.KDR-positive cells gradually increased up with the time.EPCs growth curve showed that cells proliferated fast on days 3-6.CONCLUSION:EPCs can be isolated and cultured by density gradient centrifugation from marrow suspension of SD rats.These cells exhibit the characterization of endothelial cells,and can differentiate into endothelial cells.

Objective To observe the influence of Bailong Jieyu Granules on quality of life (QOL) of patients with cancer-ralated depression. Methods A prospective and self-control clinical trial was carried out. Sixty seven patients with cancer-ralated depression were chosen and treated with Bailong Jieyu Granules. EORTC QLQ-C30 3.0 scores of patients were assessed before and after the treatment. ABSTRACT

A new algorithm, the coordinates transform iterative optimizing method based on the least square curve fitting model, is presented. This arithmetic is used for extracting the bio-impedance model parameters. It is superior to other methods, for example, its speed of the convergence is quicker, and its calculating precision is higher. The objective to extract the model parameters, such as Ri, Re, Cm and alpha, has been realized rapidly and accurately. With the aim at lowering the power consumption, decreasing the price and improving the price-to-performance ratio, a practical bio-impedance measure system with double CPUs has been built. It can be drawn from the preliminary results that the intracellular resistance Ri increased largely with an increase in working load during sitting, which reflects the ischemic change of lower limbs.

Objective:To culture and identify ratendo the lialprogen it or cells,and too bserve the expression of recombin anthuman vascular endothelial growth fator-165(VEGF165)in endothelial progenitor cells transfected with pcDNA3.1(+)/VEGF165 plasmid.Methods:Endothelial progenitor cells were cultured in vitro,and immunocytochemical staining was used to detect the expression of surface antigen(CD34,CD133,KDR).After transfection of VEGF165,VEGF protein in the culture supernatant of endothelial progenitor cells was detected by enzyme-linked immunosorbent assay(ELISA),and VEGF165 mRNA in endothelial progenitor cells was detected by reverse transcription polymerase chain reaction(RT-PCR).Results:The cultured cells were identified to be endothelial progenitor cells by immunocytochemical analysis.Gel electrophoresis showed that a 576 bp product was amplified by RT-PCR in VEGF165-transfected cells,while a weak expression of VEGF165 was observed in mock-transfected and non-transfected cells.The protein level of VEGF165 in the supernatant of VEGF165-transfected,mock-transfected and non-transfected cells were 175.8?10.7 pg/ml,10.5?1.6 pg/ml and 9.3?1.3 pg/ml,respectively.Conclusion:There is small amount of VEGF165 expression in endothelial progenitor cells in vitro,and the endothelial progenitor cells transfected with pcDNA3.1(+)/VEGF165 plasmid have an increased expression of VEGF165.
